Zusammenfassung
Explantate des Cerebrocortex neonataler Ratten wurden unter Zugabe von 5-Hydroxytryptami n.
kreatininsulfat in einer Konzentration von 0, 1 mM' bis zu 15 DIV kult iv ier t und fl uoresoenz hiato
che m isc h mittels del' Glyoxylsaure-v.heat reaction " (LL.'\DVALL und BJORKLUXD 1974) unte rsucht .
In Abhangigkeit v on K ultivabionsalter und /oder Pharmakoneinwirkungsdauer ko nnten an de n
un terschiedli chen Zell- un d Fasere lem enten in den E xpl ant ate n unterschied lich e E ffekte festgest ell t
werden. Auffiill ig ist eine R etarda t ion des \Vachstums.
Neben gelb -griine n Fluorescenzprodukten, die a ls Stoffwechselmctabolite de s Serotonin inter
pretiert werden, k ommen im Explantatzen trum Zellen mit nach Glyoxylsaure-Behandlung typischerbraun-gel ber F luorescenz VOl' .
Die Ergebnisse werden im Hinblick auf die Rolle vo n Neurotransmittern a ls ~Iodulatoren del'N eurogen ese in Analogie zu neuroembryologischen Ergebnissen diskutiert .
Summary
Explants of the cerebrocor tex of neonatal rats were cultiva ted up t o 15 DIV in addition t o 5·
hydroxytryptam ine creatinine sulfa te at the fi nal concentration of 0.1 mM .Fluorescence histochemical investigations were accomplished with a m odified glyoxylic acid
"heat reaction" ac cording to L INDVALL and B JORKLUND (1974) . D ep ending on the duration of the
cult ivat ion and/or influence of the 5-hydroxytryptamine creatinine sulfa te in varied f iber and cell
elements of the ex plants varied effec ts were found . The reta rda t ion of th e growth was st r ik ing .
Yellow-greenish fluorescence is interpreted as a fluorescence of t he metabolic substances of the
seroto nin. Cells with a brownish- yellow fluorescen ce typical of 5-hydroxytryptamine after t rea tmentwith glyoxyli c ac id in the centre of the explants were found.
Analogous to the f indings of t he neuroembryology in different species in sit u t he results were
interpreted with respect to ro le of the neurotransmitters as modulators of the neurogen esis.

aus Hirngewebe durch Al\II:-O et al. (1954) ist di e Funkt ion diesel' Substanz als Neurotransmitter und seine differen t e R olle in del' Ont ogenese des Vert ebraten-ZNS Gegen
stand vielfalt iger Untersuc hungen (BAKER und QUAY 1969).Die unterschi edli che Verteilung des 5-Hydroxytryptamills im CNS del' Vertebraten
konnte durch die Entwicklung ueuer und den kombinierten E insatz verschiedenerMethoden in den letzt en Jahron syst emat isiert werden . Besonders fluores cenzhistoehemische und kom biniert e Lasions- und neuroohemi sche E xpe rimente (SIlIIOK et a l.
1973, JACOBS et al. 1974, HALARIS et al . 1976) ha ben gezeigt, daB das gesa mte Sero
t onin des Gehirns au s Perikarya im Nucleus raphes dorsali s et medi anus mesencephalistammt (DAHLSTROM und :FUXE 1965). F ur die Funktion diesel' Substanz als Neurot ransmitter spricht u. a . die 'I'atsache, daB nach t ot aler Entleerung des Hirns vonSerotonin mittels gezielt er Lasionen im entsprechenden mesen cephalen Areal resp.in Kornbination mit drastischen pharmakologischen Eingriffen durch p-Chlorphenylalanin, a ls Tryptophan-Hydroxylase-Inhibitor, Hypersexualitat , erhoht e Aggression,SchlafJosigkeit und gesteigerte Lokomotion resultieren. Die systemat ische Applikat ion von Serot onin oder verwand te r chemischer Verbindungen zeigt offensichtlich
iiber eine Primarwirkung auf P olarisation und P ermeabilitat neuronaler Mernbranen
(WEIGHT und SALl\1OIRAGHI 1968, GROl\IOVA 1977) Effekte auf verhaltensma ffige undallgemein-physiologische Prozesse und eine vermutliche Beteiligung an del' Konsoli
dierung von Gedachtnisinha lten .Unter dem Aspekt von Induktion und Morphogenese des Vertebraten-CNS und
del' ganz offensicht lich unt erschiedli chen , alters- und somit differenzierungsabhangi,gen Ansprechbarkeit des Gehirns fiir Praekursoren und/oder Struktura naloga vonNeurotrans mit tern allgemein , die als Psychopharmaka Bedeutung besitzen, sind beidem Bernuhen , modellhafte biologische Systeme zum St udium von Transmitterwirkungen zu finden , B efunde von besonderem Interesse, die sowohl den Nachweisvon Serotonin in friihen Ontogenesestadien erbrachten als au eh eine um schriebeneWirkung exogen zugefiigter adrenerger Transmitter als Medi atoren del' friihen Ontogenese u. a. auch des Gehirns dargestellt haben (BUZNIKOV et al. 1964, 1968, 1970,1972, QUAY 1968, GUSTAFSON und TONEBY 1970, BURDEN und LAWRENCE 1973,EMANUELSON und PALEN 1975).
Fluorescenzhistochemische Untersuchungsmethoden fiir biogene Amine (FALCKund OWAN 1965, LINDVALL und BJORKLUND 1974a) haben den Vorteil gesicherterSpezifitat . Nachdem uns in vorausgeg angenen Untersuchungen die Adaptation del'
Glyoxylsaure-Technik von LmDvALL und BJORKLUND (1974a) an die histochemischn icht a ufgear beitet e Cere broco rtexkult ur gelungen (PFISTER und GOWOREK 1978 a )
und dam it ers te Studien zur Aufnahme exogenen Nora drenalins und DOPAminsmoglich waren (P FIsTER und GowoREK 1978b), haben wir nunmehr versucht, mit die
sel' Technik den EinfJuB von Seroto nin auf die Entwicklung del' Kultur zu verifizieren ,

Material und MethodenE3 wurden Explantate vom Cerebrocortex neonataler Ratten in Maximow-Doppeiglasimmmern
bis 15 DIV kultiviert. Als Stutzsubstrat dienten vorbehandelte Deckglaschen unter Verwendung
von Hahnenplasma. Nahrrnerlium: 50 % EAGLES MEM, 17 % Hiihnerembryonalextrakt, 33 %
rnenschliches Plaeentarserum; Glueosekonzentration: 600 mg/l00 ml; pH = 7,2. Dem Nahrrnediurn

Befunde
Nach Appliks t ion von 5-Hydroxytryptamin(-Serotonin-)kreat ininsulfat bei einer
Endkonzentration von 0,1 m ll'! zeigt sich im Fluorescenzmikroskop in den unter
schiedliehen Kultivationsstadien eine unterschiedliohe Morphol ogic des Explantatsund del' Waehstumszone und eine hinsichtlieh Farbe und Intensitat differente Fluo-rescenz:
1 DIV: Das Explantat ist deutli ch in ein dichtes, unspezifisch intensiv gelb fluorescierendes Zentrum und eine Verdiinnungszone gegliedert. Im E xplantatzentrum sindschemenh aft groBe, runde Zellen und wenige kleine, intensiv gelb flu orescierende,offenbar niohtneuronale Elemente zu erkennen . Von Form und GroBe denen im Explantat zentrum vergleichbar, konnen in del' Verdiinnungszone des Explantates st arkhyperplasti sehe, kreisrunde Zellen dargestellt werden, die naoh Ausbrennen des back
ground 's eine sehwaeh griinlioh-blaue Fluorescenz im Cytoplasma behalt en ; Zellkernund Zellmembran sind klar abgrenzbar (Abb. 1). Einzelne diesel' Zellelemente werden"pinocytoseartig" vom Explantat abgeschniirt (Abb, 4). Die Auswaehstumszone zeigtganz vereinzelt diinne, nieht fluoreszierende Faserelemente , jedoeh keine Zellen .
2 DIV: Das gesamte Explantat erseheint st ark aufgehellt, ein zusammenhangendes,kompakte s E xplantat zentrum ist auf einen kleinen zent ra len Teil eingeengt . Gewebegleicher Konsist enz breitet sich Iakunenartig, ausgehend vom Zentrum, noch cinekurze Streeke in Ri ehtung E xplantstrand aus , Zwischen den Lakunen von Explan tatzentrumgewebe, deutlicher jedoch in del' sicht bar breiteren Aufhellungszone sindFaserstrukturen und gelbfluoreseierende nicht-neuronale Zellelemente auszumachen(Abb. 2). Del' Erwahnung bediirfen vereinzelt vorkommende trapezoide Zellen innerhalb des Explantates. Die groBen hyperplasti schen Zellen , die bei 1 DIV fur das Explantatzentrum oharakteristisch sind und die Aufhellungszone um saumen , sind bci2 DIV nicht mehr vorhanden .
Die Faserstrukturen im Explantatzcntrum und in del' Aufhellungszone flu oresoieren grunlich. Bei 2 DIV ist eine bre itere Auswaohstumszone vorhanden und durehfolgende Strukturen chara kt erisiert : Relativ wenige diinne und verzweigte Fasern,die nach UV-E inwirkung cine griinliehe F luoreseenz beha lten und von kleinen rundenZellen mit granularer eytoplasmat isehcr Gelbfluoreseenz umsaumt sind; in die Verzweigungsstellen del' Fasern in del' Auswachstumszone sind gelegentlieh Zellen eingepallt, die cnt weder von trap ezoider Form und randate.ndiger griiner Fluoreseenz sindoder rund erseheinen und innerhalb des nieht reaktiven Cyto plasmas klumpige griineFluoreseenzprodukt e aufweisen.
4 DIV: Dem erneut kompakt erseheinenden, unspezifisch gelb flu oreseierenden Explantatzentrum st eht in diesem Ent wicklungsstadium eine ext rem breite Aufhellungs-

zone gegenuber. In letzterer imponieren Faserstrukturen mit griinlicher Fluorescenz,
die sich vereinzelt in die fluorescenzoptisch kaum auszumachende Wachstumszone
fortsetzen, Aulier' wenigen kleinen nichtneuronalen Ze11elementen in del' Aufhe11ungs
zone konnten keine fluorescierenden Ze11en gefunden werden
9 DIV: Nach 9 d Kultivation unter dem EinfluB von 5-Hydroxytryptaminkreatininsulfat ist vom Explantatzentrum ein kleiner Teil erhalten, der teilweise autolytischerscheint, in seinen kornpakten Anteilen intensiv gelb fluoresciert und vereinzeltkleine, kreisrunde, gelb-grim fluorescierende Zellelemente enthalt , Die auch in diesem
Entwieklungsst.adium sehr breite Aufhe11ungszone erseheint am Explantatrand extrern verdimnt. In der gesamten Aufhe11ungszone finden sich regelmallig geformte,
kleine, runde, schwach gelblich-grun fluoreseierende Zellen, die denen im Explantat
zentrum gleichen.
In del' Auswachstumszone konnte fluoreseenzhistoehemsch kein Befund erhobenwerden.
15 DIV: Das Explantatzentrum fluoresciert nieht massiv; es enthalt voneinander
getrennte Anhaufungen blasiger, z. T. Cytoplasmafortsatze aussendender Ze11en, indenen der Ze11kern schemenhaft bis deutlieh zu erkennen ist. Das Cytoplasma einesTeils diesel' Ze11en fluoresciert braunlich bis braunlich-gelb. Die breite Aufhe11ungszone des Explantates ist angefiillt von mehreren Reihen polygonaler Aggregate, in
denen trotz optiseh guter Verifizierbarkeit keine zellularen Strukturen erkennbar
sind. Typisch ist eine polyedrische Portionierung dieser Aggregationen dureh nichtfluoreseente, klar tingierte Spaltraume, Die Aggregate enthalten, betont am Explantatrand, feinste, granulare, blaugriine Fluoreseentprodukte, die nach Ausbrennen derUntergrundfluorescenz erhalten bleiben (Abb. 3). Gelb fluoreseierende Faserelemente
wurden sowohl im Explantatzentrum als auch zwischen den polygonalen Aggregationen der Aufhellungszone vereinzelt, abel' nieht reproduktiv beobaehtet.
In del' Auswachstumszone konnte auch in diesem Kultivationsstadium fluorescenzhistoehemiseh kein Befund erhoben werden.
Diskussion
Die gemessen am Paraformaldehyd wesentlieh hohere Kapazitat von Glyoxylsauro mit biogenen
Aminen chemisch definierte fluoreseierende Kondensate zu bilden (BJORKLU~D, LINDVALL und
SVEXSSON 1972; AXELSSON, BJORKLUND, FALCK und SVENSSON 1973) wurde von LINDVALL und
BJORKT.FXD (1974) zur Ausarboitung einer hochsensitivon fluoreseenzhistoehemisehen Methode ge
nutzt, mit del' ein distinkter Nachwci« von Noradrenalin und nOPAmin in Vibratomschmt.ton mog:
l ich ist, Auf del' Basis del' Adaptation diesel' Methode an die histotochnisch nicht aufgearbeitete Ex
plantatkultur des Cerebrocortox (PFISTER und Goworucx 1978a) interessierte nunrnehr die Frage,
ob dem 1ledium zugefiigtos Serotonin
1. im Vergleieh zu unbehandolten Kulturen Wachstums- und Differenziorungsleistung del' Ex
plantate in vitro in Abhangigkoit von dol' Kultivationsdauer und/oder Einwirkungszeit des Trans
mitters beeinflussen und
:? diese Veranderungen und das Giyoxylsauro-Kondonsat des 5-Hydroxytryptamin im F'luo
roscenzmikroskop verifizierbar sind.

Unter dem EinfluB von 5-Hydroxytryptamin kommt es im Vergleich zu unbehandelten Kulturen
zu einer eindeutigen Retardation des Wachstums und teilweisen Autolyseerscheinungen (Abb. 4).
Am deutlichsten ist diese Retardation des W'achstums an den Faserelementen erkennbar mit der
Konsequenz einer fluorescenzoptisch kaum noch auszumachenden Auswachstumszone. Die Wachs
tumshemmung betrifft aile Zellelemento bis auf einen groBzelligen Typ, der offensichtlich bis zur
viilligen Erschiipfung immer erneut aus dem Explantatzentrum nachruckt und bei 15 DIV die fur
das Serotonin-Glyoxylsaure-Kondensat typische gelb-braune Fluorescenz im Cytoplasma zeigt. Bei
diesen groBen Elementen handelt es sich so urn die einzige Zellart, die den mit dem Medium appli
zierten Transmitter akkumuliert.
Die Tatsache, daf abgeleitet von der synaptischen Lokalisation der Adenylcyclase, die Akkumula
tion von cAMP bei inkubierten Hirnsclmitten primar in neuronalen Zellen erfolgt (DE ROBERTIS
et al. 1967), konnte bei dem bekannten metabolisohen Zusammenhang von cAMP und synaptischen
Transmittern allgemein (FLOREDO et al. 1971) und uber clie adrenergen Rezeptoren (FURCHGOTT
1970) von cAMP unci Catecholaminen im besoncleren C.YlcAFEE et al. 1971, BAUDRY et al. 1976)
clarauf hindeuten, claB die in der Explantat-Aufhellungszone erscheinenclen hyperplastischen Zellen,
die bereits im Kultivationsalter von 2 DIV vollstandig autolysieren, miiglicherweise neuronale Zellen
sind. Da Kontrollversuche mit und ohne vorherige Serotoninbehandlung bei Weglassen der Glyoxyl
saureinkubation Autofluorescenzen ausschlieflen, mussen wir davon ausgehen, daB die in den Ex
plantatanteilen f1uorescierenden Produkte Glyoxylsaure-Kondensate im Ergebnis der 5-Hydroxy
tryptamin-Behandlung sind. Stoffwechselmetabolite des Serotonin sind clabei nieht auszuschlieBen,
zumal LINDVALL unci BJORKL1JND (1974a) nach Glyoxylsaure-Begaeung von mit unterschiedlichen
Aminosauren und -derivaten praparierten Proteinmatrices deren spektrale Charakteristika und Fluo
rescenzausbeute stucliert haben. Demnach karnen neben dem Serotonin selbst (relative Fluorescenz
ausbeute = 125) noch 5-Hydroxytryptophan (relative Fluorescenzausbeute = 130), N-Acetyl-5
Metoxytryptamin (relative Fluorescenzausbeute = 80) und N-Acetyl-5-Hyclroxytryptamin (relative
Fluorescenzausbeute = 60) als fluorescierende Glyoxylsaure-Kondensate mit vergleichbaren spek
tralen Charakteristika in Frage, Fur eine eincleutige Aussage uber die chemische Natur der Fluo
rescenzproclukte in unserem Untersuchungsmaterial ist eine mikrospektrofluorometrische Analyse
ebenso notwendig wie clie Arbeit mit Explantaten von in situ pharmakologisch vorbehancleltem
Tiermaterial (BLOOM uncl COSTA 1971, LIDBRINK et al. 1971, ROBERGEet al. 1972). Erfolgversprechend
durfte in diesem Zusammenhang cler Einsatz neurotoxischer Substanzen mit clem Effekt einer selektiven Degeneration von Serotoninsynapsen, -axonen unci eventuell -perikarya wie clie 6'- bzw. 7'
hydroxylierten Serotoninclcrivate sein (BAUMGARTEN et al. 1972, BAUMGARTEN 197:~, LACHEN:\1AYER
und BAUMGARTEN 1974, JACOBSet al. 1977). Es kommt hinzu, daB die bei in situ-Experimenten mit
diesen Substanzen auftretende Problematik cler unzureichenden Passage cler Blut-Hirn-Schranke,
clie bekanntermaBen fur aile Amine gilt, bei cler Explantatkultur nicht auftritt.
Vorerst konnen wir die gelb-griinlichen Fluorescenzprodukte nicht naher verifizieren. Die braun
gelbe Fluorescenz in clen Zellen cles Explantatzentrums bci 15 DIV clagegen wet-ten wir als Fluorescenz
des Kondensationsprocluktes von Serotonin, zumal LINDVALL und BJORKLUND (1974b, p. 5) fest
stellten "their fluorescence was brownish-yellow.. . ".
Trotz dieser notwendigen methoclenkritischen Einlassungen sincl nach Applikation von 5-Hyclroxy
tryptaminkreatininsulfat bei Anwendung cler Glyoxylsaure-"heat reaction" im Fluorescenzmikroskop
clie beschriebenen Effekte an Cerebrocortex-Explantaten mit eingeschrankter physiologischer Aus
sage eindeutig und reproduzierbar auszumachen. Die Ergebnisse der vorliegenclen Stuclie sprechen
dafur, claB 5-Hyclroxytryptamin an Cerebrocortex-Explantaten in vitro ahnliche Effekte (s. a. GOD
FREY et al. [1975]) hinsichtlich mcmbranassoziierter Funktions- unci Differenzierungsretarclation
hervorrufen, wie sie aus entsprechenclen auch embryologischen Experimenten in situ bei Avertebraten
unci Vertebraten bekannt sind (BUZNIKOV et al. 1964, 1968, 1970, 1972, QUAY 1968, GUSTAFSON
und TONEBY 1970, BAKE unci QUAY 1969, MEDOFF und GROSS 1971, DEEP 1972, PL1,CHINO 1972,BURDEK und LAWRENCE 1973, EMANUELSON und PALEN 1975). Genannt sei die Induktion von
